IgG fraction from rabbit polyclonal antibody obtained by immunizing rabbits with a synthetic peptide (DKTKKQAVGSPRKIQKETRK) corresponding to amino acids 521-540 of human SHPRH. The IgG fraction was purified using Protein G-Sepharose.
- Storage: Store at -20°C.
- Stability: Stable at least one year at -20°C. Avoid repeated freezing and thawing.
- Biological activity: This polyclonal antibody reacts in ELISA and Western blot (1:500 dilution) using the immunizing peptide. SHPRH is a member of the SWI/SNF family of ATPaes/helicases, it posesses a C3HC4 RING motif characteristic of ubiquitin ligase protein and it has a fundamental role in DNA repair mechanism. Upon genotoxic stress, SHPRH associates with PCNA, RAD18, and the ubiquitin-conjugating enzyme UBC(E2) and promotes methyl methanesulfonate (MMS)-induced PCNA polyubiquitation suppressing genomic instability. SHPRH gene is mutated in a number of cancer cell lines, including those from melanomas and ovarian cancer.
- Formulation: Solution at 1 mg/mL in PBS. Some of the liquid in the vial could have evaporated with changes in the final volume. However, the mass of the protein is still inside the vial.
